چکیده

LTRAVIOLET irradiation frequently induces homozygosis a t one or more loci U in heterozygous vegetative cells of Saccharomyces cerevisiae. For a particular allelic pair, the observed frequency of such inductions can approach two percent among survivors under conditions where fewer than fifty percent of the irradiated cells are killed. These alterations are almost certainly due to genetic segregation rather than to gene mutation, since: (1) complementary types (homozygous recessive and homozygous dominant) often appear as different lines of descent from a single irradiated heterozygote, (2) irradiated haploids show only rare changes from dominant to recessive, and (3) irradiated homozygous recessive cells show only rare changes to heterozygous (JAMES 1953; 1955). However, the observed homozygosis could result either from independent segregations of individual loci or from linked segregations involving whole chromosomes or parts of chromosomes. The present investigation was undertaken to obtain information on the mechanism by which the segregants are produced.

Respiratory mutant and liquid holding recovery inhibition in yeast cells

Background: Cell ability to recover from radiation damage is of great relevance in cancer treatment. It is often believed that the inhibition of cell ability to the liquid holding recovery (LHR) may be an indicator of the overall suppression of cell ability to recover from potentially lethal radiation damage. However, the literature contains no experimental evidence whether the LHR inhibition m...

Radiation-induced Dominant Lethality in Haploid and Diploid Sperm of the Wasp Mormoniella.

I N yeast, more dominant lethality is induced in diploid than in haploid vegetative cells (OWEN and MORTIMER 1956), and this is consistent with the observed increase in X-ray sensitivity of cells of higher ploidy in this organism ( MORTIMER 1958). To study dominant lethality with respect to ploidy in a higher organism, the parasitic wasp Mormoniella was selected. Stocks are available which prod...

Yeast Two Hybrid cDNA Screening of Arabidopsis thaliana for SETH4 Protein Interaction

SETH4 coding sequence with 2013 bp is a member of gene family expressed in gametophytic tissues of Arabidopsis thaliana. This fragment was PCR amplified using Kod Hi Fi DNA polymerase enzyme. This fragment was cloned into pGBKT7 bate vector and transformed E. coli DH5? cells containing vector were selected on LB medium containing Kanamycin. Finally, pGBKT7-SETH4 bate was transformed into yeast ...

YCF and YAP gene expressions in yeast cells after irradiation combined with mercury treatment

Background: All aerobically growing organisms suffer from exposure to oxidative stress, caused by partially reduced forms of molecular oxygen, known as reactive oxygen species (ROS). These are highly reactive and capable of damaging cellular constituents such as DNA, lipids and proteins. Consequently, cells from many different organisms have evolved mechanisms to protect their components...

Radio-adaptive response of peripheral blood lymphocytes following bystander effects induced by preirradiated CHO-K1 cells using the micronucleus assay

Background: Radio-adaptive response and bystander effects are known phenomena occurring in cells following exposure to ionizing radiation (IR). In this study we examined possible radio-adaptation of lymphocytes following bystander effects induced by CHO-K1 cells. Materials and Methods: Whole blood and CHO-K1 cells were cultured in RPMI-1640 complete medium. Cells were separately irradiated with...
